摘要: 本試驗目的在於探討藥用植物臺灣胡椒及石蓯蓉的最佳培養條件,建立有效的增殖系統。研究方法使用組織培養技術,誘導培植體芽體與發根的生長。
臺灣胡椒芽體增殖試驗,結果顯示以WPM培養基添加BA 0.1 mg·L-1和NAA 0.01 mg ·L-1進行增殖,八週後平均可獲得最多葉片數(芽體數) 為21個,但植株形態改變,故以BA 0.02 mg·L-1和NAA 0.002 mg·L-1的條件,平均葉片數(芽體數) 為9.8個,形態觀察與對照組比較植株發育正常,可獲最佳的芽體增殖效果;發根試驗以添加1/2 WPM加IBA 0.5 mg·L-1之發根數最多,培養三週發根率可達100 %。
石蓯蓉植株芽體增殖試驗,結果顯示以MS培養基添加BA 0.1 mg·L-1和NAA 0.01 mg ·L-1進行芽體增殖,八週後平均可獲得最多葉片數126片(芽體數12.6個);發根試驗以添加1/2 MS加IBA 0.5 mg·L-1之發根數最多,但根系形態改變,故以未添加任何生長調節劑的MS培養基,形態觀察顯示根系生長狀態良好,培養三週發根率可達100 %,可獲最佳的發根效果。
The aim of this study were researching the best culture condition of medicinal plants of Piper umbrllatum L. and Limonium sinense (Girard) Kuntze.
The research methods was tissue culture, inducing explants to shoot and root. The results indicated that in vitro micropropagation of Piper umbrllatum L. produced maximum leaves (shoots) 21 by cultivating on WPM medium supplemented with BA 0.1 mg�L-1 and NAA 0.01 mg�L-1, but the plant had a notable variations in appearance. The best shoot proliferation choice cultivates on WPM medium supplemented with BA 0.02 mg�L-1 and NAA 0.002 mg�L-1 could proliferate more leaves (shoots) 9.8 and had less mutant buds. In rooting experiment, using 1/2 WPM medium supplemented with IBA 0.5 mg�L-1 has the most roots and rooting rate as 100 % for 3 weeks.
The results indicated that in vitro micropropagation of Limonium sinense (Girard) Kuntze produced maximum leaves 126 (shoots 12.6) by cultivating on MS medium supplemented with BA 0.1 mg�L-1 and NAA 0.01 mg�L-1 was the best shoot proliferation choice. In rooting experiment, using 1/2 MS medium supplemented with IBA 0.5 mg�L-1 has the most roots, but had a notable variations in appearance. The best rooting experiment choice is MS medium had less mutant roots and rooting rate as 100 % for 3 weeks.
